Cocaine changes how genes work in brain

CHICAGO (Reuters) - Prolonged exposure to cocaine can cause permanent changes in the way genes are switched on and off in the brain, a finding that may lead to more effective treatments for many kinds of addiction, U.S. researchers said on Thursday.

A study in mice by Ian Maze of Mount Sinai School of Medicine in New York and colleagues found that chronic cocaine addiction kept a specific enzyme from doing its job of shutting off other genes in the pleasure circuits of the brain, making the mice crave the drug even more.

The study helps explain how cocaine use changes the brain, said Dr. Nora Volkow, director of the National Institute on Drug Abuse, part of the National Institutes of Health, which funded the study published in the journal Science.

"This finding is opening up our understanding about how repeated drug use modifies in long-lasting ways the function of neurons," Volkow said in a telephone interview.

For the study, the team gave one group of young mice repeated doses of cocaine and another group repeated doses of saline, then a single dose of cocaine.

They found that one way cocaine alters the reward circuits in the brain is by repressing gene 9A, which makes an enzyme that plays a critical role in switching genes on and off.

Other studies have found that animals exposed to cocaine for a long period of time undergo dramatic changes in the way certain genes are turned on and off, and they develop a strong preference for cocaine.

This study helps explain how that occurs, Volkow said, and may even lead to new ways of overcoming addiction.

In the study, Maze and colleagues showed these effects could be reversed by increasing the activity of gene 9A.

"When they do that, they completely reverse the effects of chronic cocaine use," Volkow said.

She said this mechanism is likely not confined to cocaine addiction, and could lead to a new area of addiction research for other drugs, alcohol and even nicotine addition.

"One of the questions we've had all along is, after discontinuing a drug, why do you continue to be addicted?

"This is one of the mechanisms that probably is responsible for these long-lasting modifications to the way people who are addicted to drugs perceive the world and react to it," she said.

In so doing, young gals (and their shopaholic moms, too) get to save lots of trees and keep plastic bags out of landfills.

Young ones also contribute to resource conversation by keeping in mind other green living tips like taking quick showers, buying and using products – like organic cosmetics -- that do not contain synthetic substances, as well as clothes and accessories made of organic cotton or hemp. Switching off room lights and computer equipment (including the monitor) when not in use and going for green food options are likewise some of the ways to lessen the eco footprint.

Philadelphia Public School Alumnus Gives Back to the Community -

Sponsors Youth Entrepreneur Program at the Anna Blakiston Day School

Philadelphia, PA, May 15, 2012 - Dr. Robert A. Faison is no stranger to adversity. At the age of eight he was separated from his family and lived in the Stenton Home for Children for three years. During that time, he attended the A. B. Day School where he received his primary education in a nurturing environment. When asked to sponsor a Youth Entrepreneur Program for students at his Alma Mater, he didn't hesitate.

In the past the program had been funded by the School District through The Business Center for Entrepreneurship and Social Enterprise. "When I discovered that funding wasn't available this year, I called Robert. He and I attended the University of Pitt together and I remembered that he had attended A.B. Day so I reached out to him and he accepted" said Pamela Rich-Wheeler, Director of the Business Center.

At age eleven Dr. Faison was reunited with his family. Although the adversity continued, he remained diligent in his studies, graduated from Olney High School and pursued his medical degree at the University of Pittsburgh. He then attended Yale University's School Of Medicine for Residency in Obstetrics/Gynecology (OB/GYN).

Dr. Faison had been contemplating ways to give back to his community so when presented with this opportunity he was overjoyed. On Thursday, May 17, 2012, he will come and meet the program's participants, talk with them and answer any questions they have.


The school's principal, Ms. Karen White was happy to have the opportunity to offer this after-school program to her students. Within one week 26 parents registered their children to participate. The program instructor, Kendall Hayes is impressed with the knowledge and enthusiasm of the third through sixth grade participants. "One of the first things we do is to show the students how to identify a problem and convert it into a business opportunity. These students are familiar with the basic understanding of entrepreneurship and excited about learning how to roadmap their way to success."

The program also teaches life skills, basic accounting principles (including budgeting, cash flow analysis, investing and return on investments) problem solving, the importance of a business plan, and how to write one.

Ms White says that students are very excited about meeting Dr. Faison and have lots of questions for him.
